Guidelines for method transfer in HPLC
"HPLC calculator" is a tool developed at the LCAP to improve your HPLC analytical methods in terms of analysis time and/or resolution. It is not an optimization software and will not find the mobile phase conditions necessary to separate compounds of interest. Only kinetic parameters are evaluated.
1. The calculator determines chromatographic performance (i.e. efficiencies and induced backpressures) of 9 selected chromatographic columns, from Agilent, Waters and Thermo-Electron and gives the best conditions to be used in isocratic and gradient mode, according to the selected mobile and stationary phases as well as the analyte's molecular weight.
2. Furthermore, the calculator allows to transfer isocratic or gradient LC methods thanks to fundamental equations of chromatography. For gradient transfer, the system dwell volume should be indicated for calculation.
"HPLC calculator" could be used for any transfer between HPLC systems with different column geometries, between HPLC and UHPLC (or any other sub-2µm configurations) (down-scaling), but also from analytical LC to preparative LC (up-scaling).
