Christian Lüscher, professeur au Département des neurosciences fondamentales de la Faculté de médecine de l’UNIGE et neurologue aux Hôpitaux universitaires de Genève (HUG), est le lauréat du Prix Otto Naegeli 2020 pour la recherche médicale attribué tous les deux ans à un scientifique contribuant de manière exceptionnelle à la compréhension de mécanismes biologiques ou à de nouvelles approches thérapeutiques.
Le professeur Lüscher, dont les travaux allient recherche fondamentale et translationnelle, est un spécialiste des mécanismes neuronaux de l’addiction, définie comme l'utilisation compulsive d'une drogue, même face à des conséquences négatives majeures. Il a ainsi mis en évidence les effets sur le comportement d'une libération excessive de dopamine, ce qui modifie de façon quasi permanente la transmission synaptique dans certaines régions du cerveau. Par exemple, la connexion entre le cortex et le striatum (une région sous-corticale impliquée notamment dans le mécanisme de la récompense) se renforce, expliquant pourquoi certains individus prennent des décisions pathologiques et au final développent une consommation compulsive. En d'autres termes, la récompense de la prise de drogue est surestimée tandis que ses conséquences négatives sont sous-estimées. Les personnes souffrant de toxicomanie consacrent alors toute leur énergie à une seule chose: se procurer de la drogue.
Le groupe du professeur Lüscher a en outre identifié les modifications synaptiques qui expliqueraient pourquoi certaines personnes sont susceptibles aux addictions alors que d’autres non, proposant une explicitation à une question centrale dans la recherche sur la toxicomanie. De plus, il a démontré la réversibilité du processus d’addiction dans des modèles animaux grâce à la stimulation cérébrale profonde, une technique thérapeutique déjà utilisée avec succès chez des patients souffrant de la maladie de Parkinson.
«Nous récompensons un scientifique d’exception, dont les travaux novateurs ont non seulement permis de mieux comprendre les mécanismes de l’addiction, mais aussi d’explorer de nouvelles approches thérapeutiques», indique le Comité scientifique de la Fondation Otto Naegeli. «De plus, Christian Lüscher ne s’arrête pas à la recherche fondamentale mais, en collaboration avec des neurologues cliniciens, il étudie maintenant la faisabilité de son approche chez des patients souffrant d’addiction, avec l’espoir de pouvoir offrir une stratégie thérapeutique efficace.»

Christian Lüscher, Professor at the Department of Basic Neurosciences of the UNIGE Faculty of Medicine, and Neurologist at the University Hospitals of Geneva (HUG), is the winner of the Otto Naegeli Prize for medical research 2020 , awarded every two years for an outstanding contribution to the understanding of biological mechanisms or new therapeutic approaches.
Professor Lüscher, whose work combines fundamental and translational research, is a specialist of the neural mechanisms of addiction, defined as the compulsive use of a drug, even when facing major negative consequences. He has thus highlighted the effects on behaviour of an excessive release of dopamine, which leads to long-term alterations of synaptic transmission in certain brain regions. For example, the connection between the cortex and the striatum (a subcortical region involved notably in the reward mechanism) is strengthened, explaining why some individuals develop pathological decision-making, ultimately leading to compulsive drug use. In other words, the drug reward is overestimated while the associated negative consequences are underestimated. People suffering from drug addiction then devote all their energy to one thing only: getting the drug.
Professor Lüscher's group also identified the synaptic changes that may explain why some people are susceptible to addiction while others are not, offering an explanation to a central question in addiction research. In addition, the group demonstrated the reversibility of the addiction process in animal models using deep brain stimulation, a therapy already used successfully in patients suffering from Parkinson's disease.
«We are rewarding an exceptional scientist whose innovative work has not only led to a better understanding of the mechanisms of addiction, but also to the exploration of new therapeutic approaches», says the Scientific Committee of the Otto Naegeli Foundation. «Moreover, Christian Lüscher is not stopping at basic research but, in collaboration with clinical neurologists, is now studying the feasibility of his approach in patients suffering from addiction, with the hope of offering an effective therapeutic strategy.»
