«A Molecular Basis for Pre-Formation in the Mammalian Embryo?»
The emergence and evolution of limbs was an essential step in the success of vertebrates and Hox genes are key players in this developmental process. We examined their long-range transcriptional regulation by using both chromosomal variants and approaches combining chromosome conformation capture and chromatin signatures as well as transgenic approaches in vivo. We show that a complex bimodal regulatory strategy is implemented, with two flanking regulatory landscapes somewhat pre-figuring the segmented limb morphology, thus taking us back to the old concept of preformation, yet at a molecular level. I will discuss the ontogenetic and phylogenetic rationale of this complex bimodal regulatory strategy, as well as what may control the switch between these regulatory modalities.
Biography
Denis Duboule est né en 1955. De nationalités Suisse et Française, il étudie la biologie à l’Université de Genève où il reçoit un PhD dans le domaine de l’embryologie des mammifères en 1984. Après 10 ans à l’étranger, d’abord à la faculté de Médecine de Strasbourg (CNRS), puis au Laboratoire Européen de Biologie Moléculaire, à Heidelberg (Allemagne), il est nommé Professeur ordinaire à l’Université de Genève, où il dirige le département de Génétique et Evolution depuis 1997. En 2006, il est également nommé Professeur ordinaire à l’EPFL, Lausanne, où il dirige le laboratoire de Génomique du Développement.
Ses activités de recherche s’exercent dans le domaine de l’embryologie, de la génétique et de la génomique du développement des mammifères, dans le contexte général de l’Evolution des structures et des organes. Son laboratoire a été associé à de nombreuses recherches dans le domaine de l’analyse structurelle et fonctionnelle des gènes architectes (Hox) et de leur régulation transcriptionnelle pendant le développement précoce. DD est membre de plusieurs académies et a reçu plusieurs prix et distinctions.
