OCR and ECAR rates are key indicators of mitochondrial respiration and glycolysis and these measurements provide a systems-level view of cellular metabolic function in cultured cells and ex-vivo samples. For more information on choosing the assay that is right for you

Cells are seeded into the wells of an XF cell culture microplate. During the assay, a bicarbonate-free medium should be used. The assay cartridge should be prepared before running an assay to avoid bubble formation under the sensor during the hydration.
the sensors cartridge is immersed in sterile water and placed in a CO2-free incubator at 37°C for the night.
the day of the experiment, replace water with pre-warmed XF calibrant and place the cartridge in a CO2-free incubator at 37°C for 45-60 minutes prior to load frigs into the injection port of the cartridge.
Up to 4 drugs could be added in the 4 injection ports of the cartridge, then the assay cartridge is placed into the Seahorse XF96 for calibration and then the cell plate in inserted in place of the utility plate.
